在红细胞冷冻干燥过程中,利用微CT扫描技术进行样品升华干燥过程的定时扫描观察,并对冻干品进行内部结构扫描分析,同时计算得到各个样品的孔隙率值。对冻干后的样品,采用三步法复水,用血细胞计数板进行检测。实验结果表明,有机溶剂叔丁醇可以加速冻干过程,缩短干燥时间,并且浓度为10%的比5%的显著。在升华过程中,孔隙率呈下降的趋势。最高的孔隙率出现在含有10%叔丁醇的经甘油顶处理的保护剂配方中。经过甘油预处理,细胞的恢复率得到了明显的提高,而叔丁醇的加入,导致一定程度上的恢复率下降。
In the freeze-drying process of red blood cells, the samples in plastic tube were scanned with Micro-CT scanning technique at fixed time during sublimation, the inner structures of lyophilized samples were scanning analyzed, and the porosities of freeze-drying and lyophilized samples were calculated simultaneously. The dried samples were rehydrated with three steps and evaluted by a haemacytometer. The results are as follows: Organic solvent tea-butyl alcohol (TBA) can accelerate drying process, shorten drying time, and the effect of concentration 10% is more notable than 5% one. During sublimation process, porosity show descending trend. The lyoprotective protocols containing 10% TBA and pretreated by glycerol has the highest porosity. Pretreated by glycerol is profit for cell recovery and TBA may decrease the cell recovery rate to some extent.
