探索使用甘油作保护剂提高人体红细胞冷冻干燥保存效果的可能性。甘油预处理红细胞的目的是增加细胞质浓度,减小细胞的冷冻损伤。实验分为3组:新鲜对照组、加%甘油组与40%甘油组,冻干保存前后分别检查红细胞相关指标。结果表明,20%与对照组无显著差别;40%甘油组的冻干细胞回收率与血红蛋白回收率分别维持在冷冻干燥前的55.37±4.26%和53.49±3.85%,非常显著高于对照组(P〈0.01);渗透脆性与超氧化物歧化酶(s0D)活性均显著高于对照组(P〈0.05),但与新鲜红细胞无显著差异。研究结果为使用甘油作红细胞的冻干保护剂提供了理论基础。
Pretreatment with glycerol can improve the concentration of cytoplasm of the red blood cells and then enhance the possibility of glass transition during freeze- drying process. In this research, the effects of glycerol on the preservation of freeze- dried human red blood cells were investigated. Three samples were employed in the experiment with 0%, 20%, and 40% glycerol, respectively. The results show that glycerol has remarkable effects on the cell viability. The maximum viability of red blood cells is 55.3%±4.26% after pretreatment with 40% glycerol solution. The recovery rate of hemoglobin is 53.49 ± 3.85%, which is significantly higher than that of control sample (0% glycerol). Osmotic fragility data indicate that glycerol exerts osmotic protection on RBCs during freeze - drying. The levels of superoxide dismutase( SOD) have no significant differences with that of fresh RBCs. The results provide fundamental information for long - term preservation of RBCs by freeze- drying.
