目的观察紫金龙氯仿-甲醇组分对体内急性实验性炎症的作用,探讨其抗炎作用的机制。方法于2015年4-7月,选择健康昆明小鼠60只和Wistar大鼠50只,采用醋酸致小鼠腹腔毛细血管通透性增加、角叉菜胶致大鼠足趾肿胀模型,观察紫金龙氯仿-甲醇组分的抗炎作用,并检测大鼠炎性组织中TNF-α、NO和PGE2的含量。结果与模型组相比,氯仿-甲醇组分能剂量依赖性降低小鼠腹腔渗出液的吸光度(P〈0.05,P〈0.01),中、高剂量于角叉菜胶致炎后4h分别显著降低大鼠的足趾肿胀率至31.54%和26.78%(P〈0.01),且高剂量组可显著降低大鼠炎性组织中TNF-α、NO和PGE2的含量(P〈0.05,P〈0.01)。结论紫金龙氯仿-甲醇组分具有明显的抗炎作用,其可能通过调节TNF-α、NO和PGE2炎症因子的水平,从而抑制机体的炎症反应。
Objective To observe the effect of dactylicapnos root chloroform-methanol component for internal acute experimental inflammation and study its mechanism of anti-inflammatory effect.Methods 60 healthy Kunming mice and 50 Wistar rats from April 2015 to July 2015 were selected,the permeability in abdominal cavity of mice increased caused by acetic acid,the toes of rats were swelling caused by carrageenan,and the anti-inflammatory effect of dactylicapnos root chloroform-methanol component was observed,and the contents of TNF-α,NO and PGE2 in rat inflammatory tissue were detected.Results Compared with the model group,the dactylicapnos root chloroform-methanol component could decrease the absorbance of mouse peritoneal exudatum dose-dependently,(P〈0.05,P〈0.01),after 4 h of inflammation caused by carrageenan,the middle-dose group and high-dose group respectively decreased the toe swelling rate to 31.54% and 26.78%(P〈0.01),and the high-dose group could obviously decrease the contents of TNF-α,NO and PGE2 in rat inflammatory tissue,(P〈0.05,P〈0.01).Conclusion Dactylicapnos root chloroform-methanol component has an obvious anti-inflammatory effect,and it may inhabit organismic inflammatory reactions by adjusting the TNF-α,NO and PGE2 levels.
