目的:研究跨膜激活剂及钙调亲环素配体相互作用分子的免疫球蛋白(transmembrane activator or calcium-modulating cyclophylin ligand-interactor-immunoglobulin,TACI-Ig)对rh BLy S刺激佐剂性关节炎(adjuvant arthritis,AA)大鼠肠系膜淋巴结细胞(mesenteric lymph node lymphocytes,MLNLs)功能的影响。方法:rh BLy S刺激AA大鼠MLNLs,设TACI-Ig五个浓度给药组(0.01,0.1,1.0,10,100μg/m L)和一个阴性对照组Ig G-Fc(10μg/m L)。MTT法测MLNLs增殖;流式细胞仪测T细胞主要亚群变化;ELISA法检测IL-17水平;Western blot法测TACI蛋白表达。结果:TACI-Ig降低AA大鼠MLNLs增殖能力;降低CD3^+/CD4^+和CD4^+/CD25^+T细胞,升高CD4+/CD62L+T细胞的百分含量;降低AA大鼠MLNLs中IL-17水平和TACI蛋白表达。结论:TACI-Ig对MLNLs增殖活化、细胞因子分泌及受体蛋白表达均有抑制作用。
AIM: To investigate the TACI-Ig on rh BLys stimulated MLNLs from AA rats. METHODS: MLNLs from AA rats were stimulated by rhBLy S in vitro. Set five concentrations of TACI-Ig( 0. 01,0. 1,1. 0,10,100 μg /m L) groups and a negative control group Ig G Fc( 10 μg / m L). Proliferations of MLNLs were assayed by MTT. The proportions and activity of T cell in MLNLs were analyzed by flow cytometry. The levels of IL-17 were measured by ELISA. The expressions of TACI were investigated by western blotting. RESULTS: TACIIg inhibit the proliferative reactions of MLNLs from AA rats. And TACI-Ig decreased the CD3^+CD4^+T cell and CD4^+CD25^+T cell,increased the CD4^+CD62L+T cell. Also, TACI-Ig decreased IL-17 production and TACI expression. CONCLUSION:TACI-Ig has inhibitory action on cell proliferation,cytokines secretion and receptor protein expression of MLNLs.
