目的探讨带有血小板凝血酶敏感蛋白样模体的解整链蛋白金属蛋白酶-4( ADAMTS-4)与转化生长因子β激活激酶1(TAK1)在骨关节炎(OA)软骨组织中的相关性表达。方法采用病例对照研究,分别利用免疫组织化学法和Western blot法检测ADAMTS-4、TAK1在OA组及正常对照组软骨组织中的表达情况,同时研究二者在OA中表达的相关性。结果 ADAMTS-4和TAK1在OA组表达均明显高于正常对照组( P〈0.01);二者在OA软骨组织中的表达呈正相关(r=0.469,P〈0.05)。结论 ADAMTS-4与OA软骨病变的发生发展密切相关,活化的TAK1可能致OA软骨组织中ADAMTS-4的高表达。 TAK1可以作为 OA 治疗的新靶点。
Objective To investigate the correlative expression of a disintegrin and metalloproteinase with throm-bospondin motif-4 ( ADAMTS-4 ) and TGF-activated kinase 1 ( TAK1 ) in the cartilage tissue of patients with osteo-arthritis ( OA) . Methods The case-control study, immunohistochemistry and Western blot were used to detect the expression of ADAMTS-4 and TAK1 in cartilage tissue of patients with OA and normal control group, and the ex-pressive correlation between ADAMTS-4 and TAK1 in OA was also analyzed. Results Both the expression of AD-AMTS-4 and TAK1 in OA group were significantly increased than that in normal control group ( P〈0.01 ); there was a positive correlation between the expression of ADAMTS-4 and TAK1 in OA cartilage tissue ( r=0 . 469 , P〈0.05) . Conclusion ADAMTS-4 is closely related to the occurrence and progress of OA cartilage. The activated TAK1 may cause the overexpression of ADAMTS-4 in OA cartilage tissue, which can be a new target for OA treat-ment.
