目的研究阴道超声对绒毛滋养细胞凋亡的影响及随着时间的推移其凋亡指数的变化,同时探索Bcl-2、活化Caspase-3在滋养细胞凋亡中的表达。方法收集2012年10月至2013年8月深圳市南山区蛇口人民医院收治的行人工流产前接受阴道超声照射的患者100例,设为观察组,根据阴道超声照射后流产时间的不同随机分为观察组1、观察组2、观察组3、观察组4,每组各25例。经阴道超声持续照射5分钟分别于24h、3d、5d、7d后行人工流产收集绒毛标本,另选择同期收治的未经超声照射行人工流产的患者25例设对照组。采用TUNEL法观察经阴道超声辐照后,不同天数的人胚绒毛滋养细胞的凋亡情况及随着时间的推移其凋亡指数的变化规律。同时用免疫组化SABC法检测相应滋养细胞中凋亡相关蛋白Bcl-2及活化Caspase-3表达水平的变化。结果经阴道超声照射后随取胚天数的后延,凋亡指数呈下降趋势。Bcl-2蛋白表达量逐渐增加,活化Caspase-3蛋白表达量逐渐减少。照射后1、3、5d与对照组比较差异有统计学意义(P〈0.05);但在照射后第7d与对照组比较差异无统计学意义(P〉0.05)。结论Bcl-2蛋白和活化Caspase-3蛋白在细胞凋亡转导通路中发挥了重要的调控作用,同时证实了阴道超声辐射对绒毛滋养细胞凋亡有一定的影响,但呈近期可逆性改变。
Objective To study the impact of trmasvaginal ultrasound trophoblast apoptosis of villi and its change over time, apoptotic index, while exploring the Bel - 2, Caspase - 3 activation in trophob]ast apoptosis. Methods From Octo- ber 2012 to August 2013, Shekou Peopleg Hospital of the line before abortion accept 100 patients transvaginal ultrasound irradiation, the observation group, depending on the random abortion after transvaginal ultrasound irradiation time points observation group 1 ,group 2 was observed in the observation group 3, group 4 were observed,25 cases in each group. Transvaginal ultrasound irradiation five minutes were continued for 24 hours,three days, five days, seven days after the abortion collect hair samples, another selection of patients admitted to the same period without ultrasonic irradiation in- duced abortion in 25 cases of the control group. After using the TUNEL method observed by transvaginal ultrasound irra- diation, different days of human embryonic trophohlast cell apoptosls and the variation with time of the apoptosis index. Meanwhile SABC immunohistochemistry assay nourish corresponding change of apoptosis - related proteins Bcl - 2 and activation of Caspase - 3 expression levels. Results After irradiation with transvaginal ultrasound to take a few days after the extension embryos, Bcl - 2 protein expression gradually increased, decreased activation of Caspase - 3 protein expression, 1,3,5 -day difference compared with the control group, there was irradiated significance( P 〈 0. 05 ); but in the first 7 days after irradiation with the control group, the difference was not statistically significant (P 〉 0.05 ). Conclusion Bcl -2 protein and activation of Caspase -3plays in apoptosis pathway in a very important regulatory role, but confirmed the vaginal ultrasound radiation has some in- fluence on the trophoblast cell apoptosis, but was recently reversible change.
