目的:观察兔视神经损伤后视网膜神经节细胞(retinal ganglion cell,RGC)丙二醛(malondialdehyde,MDA)/超氧化物歧化酶(superoxide dismutase,SOD)的变化。方法:选用新西兰白兔32只,分为对照组和损伤组,损伤组再根据损伤后不同时间分为3,7,14d组,每组8只16眼。采用硫代戊巴比妥酸法和黄嘌呤氧化法分别测定视网膜MDA/SOD含量。结果:损伤实验组MDA含量分别为5.95±0.78,7.67±0.64和8.29±1.02μmol/g,均明显高于正常对照组(3.82±0.54μmol/g);实验各组SOD含量分别为510±44,415±29和398±36μkat/g,均明显低于正常对照组(727±52μkat/g)。结论:自由基代谢在兔视神经损伤后RGC凋亡中起着重要作用,MDA/SOD含量测定在RGC凋亡的研究中有一定的意义。
AIM: To study the changes of MDA/SOD in retinal ganglion cell(RGC) after optic nerve injuries in rabbits. METHODS: Thirty-two New Zealand rabbits without eye diseases were divided into two groups randomly: normal control group( n = 8, 16 eyes) ; injury group( n = 24, 48 eyes). The injury group was divided into three sub groups according to different time points: 3-day group, 7-day group and 14-day group (n = 8, 16 eyes, respectively). MDA/SOD was measured respectively by TBA and xanthine oxidase. RESULTS: The content of MDA in injury groups were 5.95 ± 0. 78, 7. 67 ± 0. 64 and 8. 29 ± 1. 02μmol/g respectively, which was higher than the normal control group(3.82 ± 0.54μ mol/g). The content of SOD in injury groups were 510 ± 44, 415 ± 29 and 398 ± 36μkat/g respectively, which was lower than the normal control group (727 ± 52μ kat/g). CONCLUSION: Free radical metabolism plays an important role in RGC apoptosis after optic nerve injuries in rabbits and the determination of MDA/SOD had certain application value in the study of RGC apoptosis.
