目的观察3,6-二羟黄酮对HL-60细胞的影响。方法分别用台盼蓝拒染观察药物对细胞的毒性作用,流式细胞术、Hoechst 33258/PI荧光双染和琼脂糖凝胶电泳观察药物对细胞周期的影响以及凋亡情况。结果3,6-二羟黄酮对HL-60细胞具有明显地细胞毒作用,呈一定的时效性;流式细胞术、荧光双染和琼脂糖凝胶电泳实验,发现了典型的凋亡细胞,且随药物浓度的增加,凋亡细胞数明显增加;然而统计学分析发现3,6-二羟黄酮并没有引起HL-60细胞周期的变化。结论3,6-二羟黄酮能明显地抑制HL-60细胞的增殖,并能引起细胞的凋亡。
Objective To observe effect of 3,6-dihydroxyflavone on the proliferation of HL-60 cell lines. Methods The effect of 3,6-dihydroxyflavone on the proliferation of HL-60 cell lines was detected by Trypan blue dye exclusion assay, the changes of the cell cycle was analyzed by flow cytometry (FCM), the apoptosis was analyzed by FCM, Hoechst 33258/PI fluorescence staining and DNA agarose gel electroporesis. Results 3,6-dihydroxyflavone had strong cytotoxicity to HL-60 cell lines with dependence of time, While abnormality of cell cycles could not be induced. After treated with 3,6-dihydroxyflavone for 36h, HL-60 cell line could be induced apoptosis, which was confirmed by FCM, fluorescence staining and DNA agrose gel electroporesis. Conclusion Conclusion 3,6-dihydroxyflavone could strong inhibit the proliferation of HL-60 cell lines and could induce apoptosis.
