目的探讨microRNA-34a对宫颈癌细胞增殖及抑癌基因PTEN表达的影响。方法 Hela细胞株传代培养后分为上调组、下调组和对照组,分别转染microRNA-34a模拟物、抑制物和对照物,48 h后用CCK-8、定量PCR、Western blot法检测细胞增殖及PTEN mRNA、蛋白表达。采用生物信息软件预测microRNA-34a靶基因。结果上调组Hela细胞增殖比对照组降低23.43%(P〈0.01),而下调组比对照组增加14.77%(P〈0.01)。上调组PTEN mRNA和蛋白表达升高(P〈0.01),而下调组表达降低(P〈0.05)。靶基因预测分析显示microRNA-34a与PTEN基因mRNA的3UTR区无互补结合区。结论 microRNA-34a抑制宫颈癌细胞增殖,可能与促进抑癌基因PTEN表达有关。
Objective To investigate the effect of microRNA-34a on proliferation and PTEN gene expression of cervical cancer cells. Methods The subcultured Hela cells were divided into upregulation, downregulation and control groups, and these groups were transfected with microRNA-34a mimics, inhibitors and control agents for 48 hours, respectively. The proliferation and expression of PTEN mRNA and protein were detected by CCK-8, quantitative PCR, and Western blot, respectively. The target genes of microRNA-34a were predicted by the bioinformatics software. Results Compared with the control groups, the proliferation decreased by 23.43%(P〈0.01) but expression of PTEN mRNA and protein increased(P〈0.01) in the upregulation group. Contrarily, the proliferation increased by 14.77%(P〈0.01) but expression of PTEN mRNA and protein reduced(P〈0.05) in the downregulation group. Prediction of target genes showed no complementary binding region between microRNA-34a and 3'UTR region of PTEN mRNA. Conclusion The microRNA-34a can inhibit the proliferation of cervical cancer cells, which might be related to the reinforcement of PTEN expression.
