目的:探讨硒化壳聚糖对白血病细胞株NB4细胞VEGF mRNA表达及VEGF蛋白分泌的变化。方法:应用RT-PCR法和ELISA法研究硒化壳聚糖对NB4细胞VEGF mRNA表达及VEGF蛋白分泌的影响。结果:NB4细胞中有VEGF mRNA的表达及VEGF蛋白的分泌,硒化壳聚糖在体外能下调VEGF mRNA的表达和VEGF蛋白的分泌,呈剂量依赖关系。结论:硒化壳聚糖除可通过诱导NB4细胞分化或诱导其凋亡外,还可通过抑制促血管新生达到发挥抗白血病的效应。
Objective:TO investigate the expression of VEGF mRNA and secretion of VEGF protein in NB4 cells treated by selenium chitosan . Methods:Semi-quantitative RT-PCR and ELISA were used to study the expression of VEGF mRNA and secretion of VEGF protein in NB4 cell treated by selenium chitosan . Results:VEGF was expressed in NB4 cell. The expression of VEGF mRNA and secretion of VEGF protein could be down regulated by selenium chitosan in a dose dependent manner. Conclusion:Besides inducing apoptosis and promoting differention of Leukemia cells,selenium chitosan were exerted its anti-Leukemia effects by reducing angiogenesis via reduction of angiogenic reaction stimulating signals.
