为确定花生壳中黄酮类成分乙醇回流提取的最佳工艺条件,以黄酮得率为指标,采用响应面法对主要工艺参数进行优化并得到回归模型。方差分析结果表明:回归模型较好地反映了花生壳黄酮得率与提取时间、提取温度、乙醇体积分数和液固比的关系;最优工艺条件为提取时间2.2 h、提取温度67℃、乙醇体积分数85%、液固比13 mL/g。此工艺条件下提取花生壳黄酮得率为3.98 g/100 g,回归模型的预测值与实测值的相对误差为1.2%,该回归方程与实际情况拟合较好。
With the aim of optimization of the alcohol reflux process for extracting flavonoid from peanut hull, a central composite design (CCD) experiment was conducted and the response surface methodology (RSM) was employed to investigate the influence of four main factors including temperature, time, concentration of ethanol, ratio of solvent to raw material on the yield of flavonoid, and to establish a regression model. Regression coefficient and variance analysis show the regression model fits the actual process well. The obtained optimum extraction parameters are temperature 67℃, time 2.2 h, concentration of ethanol 85% (v/v), and ratio of solvent to raw material 13mL/g. The flavonoid yield with the optimized procedure is 3.98 g/100 g. The experimental values agree with those predicted from the regression model within a relative error of 1.2%, indicating a good fitness.
