目的利用BOOST交互作用筛选方法,查找交互作用后进行通路的发现,为疾病易感性基因的查找提供帮助。方法以WTCCC的Ⅰ期公开数据中1型糖尿病的数据为原始数据,结合BOOST方法进行交互作用分析,通过通路富集分析发现通路。结果发现了10条通路,其中有2条通路位于HLA区域外部,其余8条通路都在HLA区域内部,且通路中所包含的主要交互作用基因彼此之间存在重叠。结论利用BOOST交互作用筛选方法可有效地作为通路发现的手段,可弥补传统只考虑单变量的不足,为通路发现提供更多的可解释性。
Objective To explore the pathways of BOOST interaction screening method,and to find out the finding of disease susceptibility genes. Methods The data of type 1 diabetes mellitus in the open data of WTCCC were used as the original data,and the interaction was analyzed by BOOST method. Pathway was found by enrichment analysis. Results The results showed that 10 pathways,of which two of the pathways were outside the HLA region,and the remaining 8 pathways were within the HLA region and their containing genes were overlapped. Conclusions BOOST interaction screening method can be used as a mean for discovering susceptive pathways,which can complement the traditional methods which only consider the contributions of univariate,and also provide more explanability for the discovery.
