植物水通道蛋白(Aquaporins,AQPs)是细胞膜上选择性高效转运水分子的特异蛋白孔道.本文根据植物中hpRNA(hairpinRNA)的原理,以甘草质膜水通道蛋白基因GuPIP1为靶基因,在GuPIP1-cDNA序列中选择378bp作为构建RNAi的序列,经三次亚克隆,最后形成含GuPIP1-hpRNAi表达盒的双元表达载体,并转入农杆菌EHA105.
Aquapofins represent an important selective pathway for water to move across cellular membranes. In this study, a 378bp cDNA fragment of the Chinese licorice aquaporin GuPIP1 was chosen to construct the GuPIP1-RNAi expression cassette. The binary vector pMBWhpPIP harboring GuPIPI-hpRNAi expression cassette was constructed through three times sub-cloning via mid-clone vector and then transformed ioto Agrobocterium EHA105.
