8q24和临床相关危险因素与中国北京汉族人群前列腺癌患病风险的关系

中文摘要：

目的探索8q24（rs445114,C;rs620861,T;rs6983267,G;rs1447295,A;rs100001454,T;rs7837688,T）与中国北京汉族人群前列腺癌（prostatecancer,Pca）临床相关危险因素之间的关联性。方法收集124名Pca患者和138名正常对照者的血样和相关信息,进行基因分型,采用病例-对照和单纯病例法,进行两组间的等位基因及基因型差异分析,探讨各基因与患者的Gleason评分、PSA浓度、肿瘤分期、年龄等临床表型之间的关联。结果①8q24（rs445114,C;rs620861,T;rs6983267,G;rs1447295,A;rs100001454,T;rs7837688,T）与Pca关联分析显示各基因的风险等位基因和基因型频率在病例组和对照组间的分布差异无显著性（P〉0.05）;②单倍型分析8q24上rs445114,rs620861,rs6983267,rs1447295,rs100001454,rs7837688的单倍型分析,6种单倍型在病例和对照中均无显著性差异（P〉0.05）;③基因环境交互作用分析：携带8q24（rs445114,C）TC基因型以及8q24（rs620861,T）TC基因型的人群Pca易感性与食用洋葱负相关（P1=0.007,OR=0.31,95%CI=0.12-0.79;P2=0.005,OR=0.30,95%CI=0.11-0.77）,携带8q24（rs6983267,G）TG基因型的人群Pca易感性与饮酒正相关（P=0.007,OR=10.75,95%CI=1.35～86.14）,其他位点与Pca的发病年龄、Gleason评分、PSA浓度以及肿瘤分期等指标均无显著相关性（P〉0.05）。结论中国北京汉族人群中携带8q24（rs445114,C）TC基因型以及8q24（rs620861,T）TC基因型的人群Pca易感性与食用洋葱负相关,携带8q24（rs6983267,G）TG基因型的人群Pca易感性与饮酒正相关.

英文摘要：

Objectives To explore whether 8q24 （rs445114,C;rs620861,T;rs6983267,G;rs1447295,A;rs100001454,T;rs7837688,T） have the association with Pca and the clinic risk factor in Chinese Han populations.Method：Collect blood and information of 124 Pca patients and 138 controls.Type gene,set up gene data-base and analyze the association of Chinese Pca in the case-control and case-only model through the way of nonparametric.Analyze the interaction between genes and the environment.Explore the link between Gleason PSA,the tumor points,age,etc.with clinical characteristics.Result ①There is no remarkable different distribution of risk alleles and genotype frequencies in cases and controls among the association of 8q24（rs445114,C;rs620861,T;rs6983267,G;rs1447295,A;rs100001454,T;rs7837688,T） and Pca（P0.05）.②There is no remarkable different distribution among the haplotype of rs445114,rs620861,rs6983267,rs1447295,rs100001454,rs7837688 in 8q24（P0.05）.③the interaction between genes and the environment people with genotype TC on the site of 8q24（rs445114,C） and people with genotype TC on the site of 8q24（rs620861,T） have less probability to get Pca with onion（P1=0.007,OR=0.31,95%CI=0.12～0.79;P2=0.005,OR=0.30,95%CI=0.11～0.77）.People with genitype TG on the site of 8q24（rs6983267,G） have more probability to get Pca with alcohol（P=0.007,OR=10.75,95%CI=1.35～86.14）.Other sites are no significant relevance with Pca the age grading,psa level,Gleason and pathology on indicators （P0.05）.Conclusion People with genotype TC on the site of rs445114 and people with genotype TC on the site of rs620861 have less probability to get Pca with onion;people with genitype TG on the site of rs6983267 have more probability to get Pca with alcohol.
