为了提高高原藏猪人工授精的成功率,试验采用猪精液质量自动检测系统对8℃、17℃、25℃不同保存温度的藏猪精液进行质量检测,并统计活精子数及pH值。结果表明:在8℃、17℃、25℃条件下藏猪精液色泽无明显变化;在第72小时,25℃条件下精液散发出浓烈的腥臭味;第12,24小时,8℃、17℃、25℃温度下精液的活精子数无显著变化(P〉0.05);第36小时,在17℃和25℃下活精子数显著高于8℃下保存的精液(P〈0.05);第96小时,17℃保存的精液活精子数明显高于25℃,差异不显著(P〉0.05);17℃保存条件下,精液pH值第12小时下降了0.09,第24小时后上升到7.31,第60小时达到7.47,之后趋于稳定。说明17℃的保存温度是藏猪精液长时间保存的最佳选择,精子各项指标最佳;藏猪精液pH值总体呈现先下降后上升,最后趋于稳定的趋势,在pH值呈弱碱性条件下对藏猪精液稀释液进行短期的保存是可行的。
To improve the successful rate of artificial insemination of Tibet pig,the Tibet pig semen quality at three different storage temperatures,8 ℃,17 ℃ and 25 ℃,was detected using the automatic detection system. And then the live sperm number and pH value were counted.The results showed that the color of boar semen did not show obvious changes under three preservation temperatures. The semen stored at 25 ℃exudes strong fishy smell after 72 h. But there were no significant difference between the number of living semen under three temperatures( P 〉0. 05) at 12 h and 24 h. The number of living semen stored at 17 ℃ and 25 ℃ were significantly higher than that at 8 ℃ after 36 h( P 〈0. 05). The number of living semen stored at 17 ℃ was higher than that at 25 ℃ after 96 h and the difference was not significant( P 〉 0. 05).When stored at 17 ℃,the semen pH value decreased to 0. 09 after 12 h,then rose up to 7. 31 after 24 h and reached to the peak at 7. 47 after60 h,then tended to be basically stable. Therefore,the best storage temperature for the longest preservation of the Tibet pig semen was 17 ℃at which all the indicators were at the best performance. The pH value showed a decreased at first,then increased and finally to be stable.These indicated that the weakly alkaline pH conditions was feasible for short-term preservation of the Tibet pig semen.
