以大球盖菇为原料,通过正交试验IJ9(3。)研究超声波提取多糖的工艺条件。结果表明,最佳提取工艺条件为提取温度65℃、提取时间1.0h、超声波功率600w、料液比1:35(g/mL),此条件下,大球盖菇多糖得率为8.16%。提取效果影响大小的先后顺序为提取时间〉提取温度〉提取功率〉料液比。应用化学发光法对大球盖菇粗多糖的清除-OH和·O2自由基的能力进行了研究,结果表明:大球盖菇粗多糖对·OH和·O2^-自由基均具有明显的清除能力,清除·O2^-自由基的IC50为108.60μg/mL,清除·OH自由基的Ic50为345.98μg/mL,大球盖菇粗多糖对·0:_自由基的清除能力是对·OH自由基的清除能力的3.2倍。
In this study, the process conditions for extracting polysaccharides from Stropharia rugosoannulata with the assistance of ultrasound were optimized by L9(34) orthogonal array design methods. The optimal extraction process conditions were determined as follows: extraction temperature, 65 ℃ ; ultrasound power, 600W; and material/liquid ratio, 1 : 35 (g/mL) for an extraction duration of 1.0 h. Under these conditions, the extraction yield of polysaccharides from Stropharia rugosoannulata was 8.16 %. The decreasing order of importance of the four process conditions affecting polysaccharide extraction was extraction duration, extraction temperature, ultrasound power and material/liquid ratio. The scavenging ability of hydroxyl radicals and superoxide anion radicals of Strophctria rugosoannulata polysaccharide were assayed by chemiluminescence.There suits showed that Stropharia rugosoannulata polysaccharide have evident abilities of scavenging hydroxyl radicals and superoxide anion radicals.The crude Stropharia rugosoannulata polysaccharides obtained had strong superoxide anion radicals scavenging effect, with an IC50 of 108.60 μg/mL, which was more than 3.2-fold increased after hydroxyl radicals, revealing an IC50 of 345.98 μg/mL.
