目的研究皮肤急性光损伤、慢性光老化Cathepsin B的改变情况。方法 30块小儿包皮标本,将其随机均分为急性光损伤组、慢性光老化组和空白对照组,每组各10例。经诱导、SA-β-Gal染色、Western印迹检测、RT-PCR检测,对三组Cathepsin B蛋白条带灰度值、Cathepsin B mRNAΔCt值及Cathepsin B活性进行对比分析。结果急性光损伤组Cathepsin B灰度值高于慢性光老化组,但不具统计学差异(P〉0.05),急性光损伤组、慢性光老化组与空白对照组相较,均呈极显著差异(P〈0.01)。急性光损伤组Cathepsin B mRNAΔCt值与慢性光老化组相较,呈极显著性差异(P〈0.01),与空白对照组相较,不具差异(P〉0.05)。慢性光老化组Cathepsin B mRNAΔCt值与空白对照组相较,呈极显著性差异(P〈0.01)。急性光损伤组Cathepsin B活性与慢性光老化组、空白对照组相较,均呈显著性差异(P〈0.01),慢性光老化组Cathepsin B活性与空白对照组相较,呈极显著性差异(P〈0.01)。结论皮肤急性光损伤、慢性光老化均会使Cathepsin B表达水平降低,但急性光损伤时,Cathepsin B活性损伤程度等指标,均明显低于慢性光老化,提示急性光损伤的自我修复能力优于慢性光老化,临床应辨别区分两者异同,给予合理方案。
Objective To research the situation change of acute light damage,chronic skin photoaging Cathepsin B. Methods Selecting 30 blocks of pediatric prepuce specimens,and randomly dividing them into acute light damage group( n = 10),chronic light aging group( n = 10) and blank control group( n = 10). Induction,SA- beta- Gal staining,Western blotting,RT- PCR detection of three groups of Cathepsin,B protein bands of gray value,Cathepsin B mRNA Delta Ct value and Cathepsin B activity were compared and analyzed. Results Acute light damage group Cathepsin B gray value is higher than the chronic light aging group,but with no statistical difference( P 0. 05). In acute light damage group,chronic light aging group and blank control group,in contrast,there was significant difference( P 0. 01). Through comparing Cathepsin B mRNA ΔCt in acute light damage group and chronic light aging group,there were significantly differences( P 0. 01),and there was no difference compared with blank control group( P 0. 05). Through comparing Cathepsin B mRNA Delta Ct value in chronic light aging group with the blank control group,there was significant difference( P 0. 01).Cathepsin B activity in acute light damage in acute injury group,chronic light aging group and blank control group showed the significant difference( P 0. 01). Through comparing Cathepsin B activity in chronic light aging group and blank control group,there were significantly differences( P 0. 01). Conclusion Acute light damage and chronic skin light aging will make Cathepsin B expression level decrease.But in the acute light damage,Cathepsin B activity of damage degree was significantly lower than that of chronic light aging,which hints acute light damage self repair ability is superior to chronic light aging. The similarities and differences between the two should be judged in clinic,and a reasonable solution should be given.
