目的应用相关软件和在线网络分析Em95的蛋白二级结构,预测B细胞表位和T细胞表位的联合表位。方法采用DNAStar软件和在线网络IEDB、SYFPEITHI及Propred等对Em95的B细胞表位和T细胞表位进行预测,寻找B细胞和T细胞表位共同的区域。结果 Em95存在潜在的抗原表位,其中B细胞表位区域为10-19、43-48、52-59、78-82、92-100和110-115;分值高的T细胞表位区域为32-40、51-60、82-98、108-116和126-138。B细胞和T细胞共同的表位区域为52-59、92-98和110-115。结论运用生物信息学方法确定Em95的3个T-B细胞联合表位存在于5个T细胞抗原表位和6个B细胞抗原表位中存在区域,对进一步研究Em95的抗原性和研发优势表位疫苗具有重要意义。
Objective In order to predict combined B-and T-cell epitopes,the protein secondary structure of the Em95 antigen was analyzed using related software and the Internet.Methods To look for the common regions of B-and T-cell epitopes of Em9,the B-and T-cell epitopes of Em95 were predicted using DNAStar and IEDB,SYFPEITHI,and Propred online.Results Em95 has many latent antigenic epitopes.The possible B cell epitopes of Em95 are in the regions of 10-19,43-48,52-59,78-82,92-100,and 110-115.High-scoring T-cell epitopes are in the regions of 32-40,51-60,82-98,108-116,and 126-138.The common regions of B-and T-cell epitopes of Em95 are 52-59,92-98,and 110-115.Conclusion Bioinformatics indicated that there are three combined B-and T-cell epitopes in Em95,with 6 B-cell antigenic epitopes and 5 T-cell antigenic epitopes.These results are vital to further research on the antigenic characteristics and development of vaccines containing the dominant epitopes of Em95.
