目的:观察参竹精片对骨质疏松症大鼠胫骨密度及病理形态的影响。方法 :采用手术摘除大鼠双侧卵巢建立骨质疏松症模型,用参竹精片灌胃治疗30天,测量胫骨重量/体积比、骨密度(BV)、骨小梁数量(TB.N)、骨小梁厚度(Tb.Th)、骨小梁分离度(Tb.Sp)及骨连接密度(Conn.D);测定血清骨钙素(OC)和ALP浓度;观察胫骨病理变化。结果:参竹精片360 mg/kg、180mg/kg、90mg/kg三个剂量治疗组大鼠胫骨重量/体积比均明显增加(模型组,1.36±0.07 g/cm~3;参竹精片分别为1.65±0.07g/cm~3,1.61±0.06g/cm~3,1.65±0.06 g/cm~3;BV明显增加(模型组,933.88±8.97 mg AHccm;参竹精片360 mg/kg、90mg/kg剂量组分别为959.29±5.76 mg AHccm和966.39±5.51mg AHccm;血清OC浓度明显增加(模型组205.91±11.46 pg/ml;参竹精片180mg/kg、90mg/kg剂量组分别为308.40±47.80 pg/ml和350.32±45.25 pg/ml;ALP浓度降低(模型组55.79±12.42 ng/ml;参竹精片360mg/kg、90mg/kg剂量组分别为41.34±12.50 ng/ml和40.89±12.92ng/ml;胫骨骨质结构紧密,密度增加,裂纹明显减少。结论:参竹精片可增加骨密度,改善骨结构,对骨质疏松症具有一定治疗作用。
Objective: To explore the effect of Shenzujing tablets on tibia of SD rats with osteoporosis in bone mineral density and pathomorphism.Methods: the osteoporosis( OP) rat was established by the method of ovariectomy.50 of rats with established osteoporosis were randomly divided into 5 groups: model group,estradiol( E2)( 15mg/kg) group and Shenzujing tablets( 0.36,0.18 g,0.09g/kg) groups.Each group contained ten rats.10 healthy mice were selected as normal group.The rats with established osteoporosis received daily oral drugs for 30 days.The normal group and the model group received daily oral isovolumic distilled water for 30 days.After 30 days’ treatment,The ratio of weight and volume in shin bone,bone mineral density( BV),trabecular number( Tb.N),trabecular thickness( Tb.Th),trabecular separation( Tb.Sp),connection density( Conn.D) were measured.The contents of estradiol( E2) and osteogenin( OC) and alkaline phosphatase( ALP) in serum were detected by enzyme-linked immunosorbent assay( ELISA).The proximal tibial metaphysis were obtained for histopathological examination.We observed the pathological changes in the tibia.Results: after 30 days’ treatment,compared with the model group,the ratio of weight and volume in shin bone in Shenzujing tablets( 0.36,0.18,0.09g/kg) groups significantly increased( P 〈 0.05).BV in Shenzujing tablets( 0.36,0.09g/kg) groups significantly increased( P 〈 0.05).The contents of osteogenin( OC) in serum in shenzujing tablets( 0.18,0.09g/kg) groups significantly increased( P 〈 0.01).The contents of alkaline phosphatase( ALP) in serum in Shenzujing tablets( 0.36,0.09g/kg) groups significantly decreased( P 〈 0.05).The tibia bone structure was tight,density increased,cracks decreased significantly.Conclusion: Shenzujing tablets can increase the contents of osteogenin( OC) in serum and the ratio of weight and volume in shin bone,upregulate the bone mineral density in SD rats
