2型糖尿病正成为日益突出的世界性健康问题。胰岛素抵抗在2型糖尿病的发病中起到了重要的促进作用。近年来,很多研究发现,内质网应激与胰岛素抵抗具有密切的联系。内质网应激不仅是胰岛素信号通路的直接负调节因素,还可通过多种方式作用于不同靶组织促进胰岛素抵抗。该文就内质网应激与胰岛素抵抗在2型糖尿病发病中的作用机制及可能的靶向治疗策略作一综述。
Type 2 diabetes mellitus (T2DM) is increasingly becoming a prominent global health problem Insulin resistance (IR) plays an important role in promoting the development of T2DM. In recent years, a bunch of studies find that the endoplasmic reticulum stress (ER stress) and IR are inseparable. ER stress acts not only as the direct negative regulatory factor in the insulin signaling pathway, but also targets different tissues in various ways to enhance systemic insulin resistance. In this review, we will give a brief review about the molecular mechanism of ER stress and IR in the occurrence and development of T2DM, as well as the possible targeted therapeutic strategy toward it.
