目的探讨法呢醇X受体(FXR)与瘦素(Lep)在胆管癌组织中的表达变化及意义,并分析二者之间的关系。方法采用免疫组织化学染色法检测胆管癌组、癌旁胆管组和手术边缘正常胆管组中FXR和Lep的表达,比较各组中两者表达的差异性。结果 FXR和Lep在胆管癌组、癌旁胆管组和手术边缘正常胆管组中均有表达。FXR在癌旁无瘤切缘和正常胆管组织中的表达显著高于癌组织(P〈0.05),Lep在癌旁无瘤切缘和正常胆管组织中的表达显著低于癌组织(P〈0.05),而二者在癌旁无瘤切缘与正常胆管组织中表达均无显著差异(P〉0.05)。FXR和Lep具有相关性,相关系数为r=0.627 7(P〈0.05)。结论 FXR与Lep可能通过调节脂质代谢平衡及免疫应答等机制,共同参与胆管癌的发生和发展。
Objective To study the expression and role of FXR and Lep in Cholangiocarcinoma and the relationship between them. Methods The expressions of FXR and Lep were determined by immunohistochemistry in cholangiocarcinoma tissues,normal bile duct tissues tumor and adjacent histologically noncancerous bile duct tissues. And the differences of the two groups were compared. Results The expressions of FXR in normal bile duct tissues tumor and adjacent histologically noncancerous bile duct tissues were higher than in cholangiocarcinoma tissues( P 〈0. 05). The expressions of Lep in normal bile duct tissues tumor and adjacent histologically noncancerous bile duct tissues were lower than in cholangiocarcinoma tissues( P〈 0. 05). The expressions of FXR and Lep were not significantly different in carcinomas and paracancerous tissues of the bile duct. There was correlation between FXR and Lep,r = 0. 627 7( P 〈0. 05). Conclusion FXR and lep were mostly likely involved in the development and progression of cholangiocarcinoma by adjusting lipid metabolism and immune response mechanism.
