对古尼拟青霉小孢变种RCEF0864的深层发酵产物进行了有效成分分析,结果表明,菌丝体和发酵液中氨基酸总量明显高于冬虫夏草生药;菌丝体中粗蛋白、总糖、麦角甾醇、甘露醇及腺苷等多种物质的含量亦高于冬虫夏草生药;RCEF0864深层发酵菌丝体和发酵液中均含有丰富的对人体健康非常重要的Zn、Fe、Ca等元素,尤以Ca的含量最高;用二苯代苦味肼基自由基(DPPH)-TLC法和酶标仪法对RCEF0864菌丝体和发酵液冻干粉甲醇提取物中的清除自由基活性物质进行了分析,发现2种提取物均具有较强的清除自由基活性,在浓度为5.0mg/mL,37℃下保温10min时,2种提取物对0.4mg/mL的DPPH自由基的清除率分别为53.86%和59.66%。
The analysis on effective components in the fermentation mycelia and broth of Paecilomyces gunnii var. minor RCEF0864 showed that the contents of aminoacids, raw protein, polysaccharide, ergosterol, adenosine were higher than those in Cordyceps sinensis. Some important metallic elements such as Zn, Fe, Ca. etc. were richer in the fermentation mycelia and broth of P. gunnii var. minor RCEF0864. A DPPH-TLC and DPPH-Mcroplate assay were made to determine the free radical scavenging activity of methanol extract from fermentation mycelia and broth of RCEF0864. The results revealed that the two kinds of extracts has strong free radical scavenging activity. At the concentration of 5.0 mg/mL they can respect a decrease of 53.86% and 59.66% 0.4 mg/mL DPPH radicals after incubated at 37℃ for 10 minutes.
