蝉拟青霉高产虫草素菌株液体培养工艺的研究

为提高蝉拟青霉RCEF1081虫草素的代谢量,以虫草素含量为检测指标,用高效液相色谱法作为检测方法,通过正交试验对其高产虫草素的液体培养工艺进行优化.结果表明菌株RCEF1081最佳液体培养基配方为:麸皮3%,酵母粉4%,丝氨酸0.15%,KH2PO4 0.05%,Mg—SO4·7H2O 0.01%,CaCl2 0.02%;最佳培养条件为:起始PH值6.0,装液量80/250mL(V/V),接种量5%(V/V),培养温度25℃,摇床转速130r/min,培养时间4d,在此条件下培养其虫草素含量最高,可达2.7720mg/g,比优化前提高了144%,经统计学分析,差异极显著(P〈0.01).

英文摘要:

In order to improve the metabolism yield of cordycepin in the strains of Paecilomyces cicadae RCEF1081,we studied the liquid culture technology of it, through determining the contents of cordycepin in the mycelia of RCEF1081 by HPLC. The results showed that the optimum medium components were: bran3%,yeast 4%,serine 0.15%,KH2PO4 0.05%, MgSO4 · 7H2O 0.01%, CaCl2 0.02%; the optimum culture conditions were: initial pH 6.0, culture medium amount 80/250 ml (V/V) flask, inoculum 5% (V/V) , culture temperature 25℃, shake rotation 130 r/min, fermentation period 4 day, the total yield of cordycepin could reach as high as 2. 7720mg/g and was raised 144% compared with that of before. Through the statistical analysis, we can know the difference is significant (P〈0.01).
