应用SRAP(Sequence Related Amplified Polymorphism)标记,通过对6份待检测杂交油菜种子样品(油研50杂交种)和4份杂交油菜对照样品(油研9号、油研10号、油研11号和油研50)进行遗传多态性分析,计算出10份样品间的遗传相似性系数并绘制聚类分析图。以相似性系数的大小来鉴定6份待测样品的品种真实性。本研究为种子管理部门针对油菜种子执法提供了一种鉴定杂交种真实性的快速方法。结果表明:对10份样品进行SRAP引物组合筛选,在分析的80对引物组合中找到多态性引物组合39对,检测到多态性位点101个。经聚类分析表明,6份待鉴样品和对照样品油研50被聚为一类,它们之间的相似性系数在0.92~0.96之间,均大于0.9,而油研9号、油研10号、油研11号被聚为其它3类,它们与油研50之间相似性系数在0.37-0.48之间,均小于0.9。因此本研究认为,以相似性系数0.9为阈值是一种鉴定油菜杂交种真实性的可行办法。
Sequence Related Amplified Polymorphism (SRAP) was applied to analyze the genetic diversity of 6 rapeseed hybrid seed samples ( Youyan 50) and 4 check samples, including Youyan No. 9, Youyan No. 10, Youyan No. 11 and Youyan 50. The genetic similarity coefficients (GSC) were calculated and the dendrogram was drawn. The identification of 6 Youyan 50 samples was evaluated by the GSCs. The results would help the seed administration department identify the rapeseed hybrid seed rapidly. Results showed that 39 polymorphic SRAP primer combinations were found in the total 80 primer combinations. All these polymorphic SRAP primers produced 101 clear bands. By UPGMA cluster analysis,the 6 Youyan 50 samples and the Youyan 50 check were clustered into one group and the GSCs varied from 0.92 to 0.96. The other three samples including Youyan No. 9, Youyan No. 10 and Youyan No. 11 were distinguished as three other different groups and the GSCs between them and Youyan 50 check ranged from 0.37 to 0.48, greatly less than 0.9. Therefore, the GSC of 0.9 could be used as a threshold to identify the rapeseed hybrid seed.
