目的探究WNT5a在小鼠支气管哮喘模型肺组织中的表达。方法将24只脱敏饮食3周的6周龄Balb/c小鼠随机分为2组:对照组和哮喘组。哮喘组小鼠在第0、14天分别腹腔注射0.2 ml致敏液[100μg OVA,100μl PBS,100μl Al(OH)3凝胶]致敏,于第21-30天1%OVA雾化。对照组小鼠在同时间给予同剂量的PBS。于最后1次处理后masson染色观察小鼠肺组织上皮下纤维化的情况,QT-PCR检测小鼠肺匀浆中Wnt5a、TGF-β1的分子表达,Western blot检测肺匀浆中Wnt5a的蛋白表达。结果哮喘组小鼠上皮下纤维化增多,TGF-β1表达增加;肺组织中Wnt5a较对照组表达增加。结论小鼠哮喘模型肺匀浆中Wnt5a在分子和蛋白水平表达较对照组明显增加,这可能和哮喘中气道重塑相关。
To evaluate the expression of Wnt5 a in the asthma mouse model, 24 six-week-old female Babl/c mice were randomly divided into two groups. OVA was injected by intraperitoneal injection to set up the asthma model. After successfully established the asthma model, the subepithelial fibrosis were detected by masson staining and QT-PCR, and the expression of Wnt5 a and TGF-β1 in pulmonary homogenate were detected via QT-PCR and Western blot. Data showed the expression of Wnt5 a was elevated in the asthma mice. In conclusion, the Wnt5 a signaling pathway is activated in asthmatic mice mode, which may related with airway remodeling.
