生物转化液中大量菌体细胞、蛋白、细胞碎片的存在导致产物回收困难,本文考察了运用混凝技术、破乳技术去除转化液中的菌体及蛋白质的可行性.实验结果揭示,添加铝盐、铁盐及其高聚物有助于去除转化液中的菌体及蛋白质;对于已发生乳化现象的转化液,则需要添加少量表面活性剂如十二烷基硫酸钠后再进行混凝处理.处理过的生物转化液,蛋白质、菌体去除率接近100%,方便后续产物回收.工作为工业生物催化过程下游产品回收提供借鉴.
The presence of cells, proteins and cell debris in bioconversion mixtures makes it diffiruinous, ferrous coagulants caused coagulation of ceils and proteins. Regarding the emulsified bioconversion mixtures, the coagulation occurred through introducing a small amount of surfactant sodium dodecyl sulfate, followed by coagulant treatment. Result demonstrated that the developed pretreatment process could remove nearly 100% of cells and proteins, beneficial to product recovery improvement. This work provided bio-industry valuable reference data for downstream bioprocessing.
