聚丙烯酰胺凝胶柱分离制备高纯度纤维低聚糖的研究

利用聚丙烯酰胺凝胶柱可分离制备纤维低聚糖。在其它分离条件相同、洗脱速度不同的情况下,完成一次分离需要的洗脱剂用量是不变的,洗脱剂用量对低聚糖的分离没有影响。采用一次分离技术可得到纯度分别为96.67%、68.76%和82.00%的纤维五糖、四糖和三糖组分,纤维低聚糖混合物的回收率为81.38%。将多次分离得到的各低聚糖组分在相同的分离条件下进行二次分离,五糖、四糖和三糖的纯度分别提高到98.45%、98.65%和98.12%,回收率分别为90.91%、47.42%和68.68%。

英文摘要:

A procedure for preparative separation of purified cellooligosaccharides was developed by polyacrylamide gel column. Under similar conditions, the dosage of eluent for finishing a one-step separation process did not change with the eluting rate, The dosage of eluent had no effect on the separation. The purities of fractions of eellopentaose, eellotetraose and eellotriose separated by one-step process were 96.67 % , 68.76 % and 82. O0 % , respectively. The overall recovery of eellooligosaeeharides mixture was 81.38 %. High-purity cellooligosaceharides were obtained with purity of 98.45 % for eellopentaose, 98.65 % for eellotetraose and 98.12 % for eellotriose by the second separation under the same separating conditions. The corresponding recoveries for eellopentaose, cellotetraose and eellotriose were 90.91% , 47.42 % and 68.68 %, respectively.
