目的:探讨妊娠酒精暴露对仔鼠眼球和视皮质发育的影响。方法:妊娠母鼠经酒精灌胃法制作胎儿酒精综合征模型,检查新生仔鼠发育情况,并对1例双侧畸形眼球行HE和Nissl染色,观察眼球的组织学改变,同时用DiI散射标记视皮质锥体细胞,分析椎体细胞侧树突表面的树突棘改变。结果:畸形眼球变小,眼裂消失,角膜、晶状体和玻璃体完全缺失;视网膜片层状构筑紊乱,细胞增殖,增殖的细胞侵入眼球内腔,几乎占据整个眼球腔位置;视皮质锥体细胞树突棘变短,变稀疏。结论:妊娠酒精暴露可致眼球畸形,其原因可能是酒精毒性造成的基因突变引发的。由于发育缺陷的眼球对光反应减弱,造成视皮质锥体细胞树突棘的萎缩。
Objective:To investigate the prenatal alcohol exposure effects to the development of eyeballs and visual cortices of mice.Methods:Fetal alcohol syndrome animal model was made through intragastric gavage,and the development of teratic mice were examined.One pair teratic eyeballs were performed with H.E.staining and Nissl method,in order to observe their pathologic alterations.In the meantime,DiI diolistic assay was carried out to label the dendritic spines of pyramidal cells in visual cortices.Results:Teratic eyeballs became small,their palpebral fissures were closed,the corneas,lenses and vitreous bodies were absent completely.Furthermore,the laminar structure of retina was out of order,and the cells in retina proliferated.The proliferated cells invaded into and occupied the cavity of eyeball.In the visual cortex,the dendritic spine of pyramidal cell became short and sparse.Conclusion:Prenatal alcohol exposure would induce malformation of eyeball in mice.The gene mutation probably was the cause of teratogenesis which was induced by alcohol's toxicity.Due to the weak reaction to light in teratic eyeball,the dendritic spine in pyramidal cell of visual cortex would wither.
