衰老研究是近来国内外的一个研究热点,酿酒酵母(Saccharomy cescerevisiae)作为低等单细胞真核生物,其寿命有复制型寿命和时序型寿命之分,相对哺乳动物来说可以便捷地进行寿命测定,使得酿酒酵母成为寿命和衰老研究的模式生物。以基因同源重组技术为基础构建酿酒酵母突变菌株,检测酵母细胞的复制型寿命和时序型寿命是目前寿命研究中的经典方法。本文着重介绍酵母突变菌株的构建方法,包括单基因缺失、双基因缺失和基因过表达;酵母的复制型寿命、时序型寿命的检测方法;酵母寿命调节机制的研究进展。
Aging research is a hot topic of life science. Replicative life span and chronological life span of Saecharomy cescerevi- siae can be tested easily, which makes yeast to be an ideal model organism in life span and aging research. Classic methods of stud- ying yeast life span are to detect replicative life span and chronological life span of mutant cells that constructed by homologous recom- bination. In this paper construction methods of yeast mutant strains are recommended including mono or dual gene knockout and gene overexpression strains. How to detect the replicative life span and chronological life span of yeast is discussed. The life span - regula- tory pathways are also introduced in this paper.
