以提取量为考察指标,优化超声波辅助提取葡萄果皮色素的工艺。在单因素试验基础上,选取乙醇浓度、超声处理温度、超声处理功率为主要影响因素,进行3因素3水平的Box-Behnken中心组合试验设计,以葡萄果皮色素提取量为响应值进行响应面分析,优化超声波辅助提取色素的条件。结果表明:超声波辅助提取色素的最佳工艺参数为:乙醇浓度39.49%,液料比为20∶1,超声处理温度65℃,超声处理功率为最大功率的60.81%,浸提时间20min;在最佳提取条件下,色素提取量理论值为26.29,验证值为26.00,二者之间的相对误差为1.09%。
Pigments in grape skin were extracted with the assistance of ultrasound.The effects of ethanol concentration,ultrasound temperature,and ultrasound power on the yield of pigments were studied by conducting single factor tests,and Box-Behnken experimental design involving the 3factors at 3levels was employed.In order to optimize conditions for extracting grape pigments from grape skin,response surface methodology was used.The results indicated that the optimum extraction parameters are as follows:ethanol concentration of 39.5%,liquid-to-solid ratio of 20∶1,at 65℃,ultrasound power of 60.81% of the maximum and extraction for 20 min.Under thoes conditions,the theoretical yield of pigments is 26.29,while the experimental value is 26.0,and their relative stand error is 1.09%.
