RASSF1A和p16 mRNA在非小细胞肺癌组织中的表达及意义

目的探讨RASSF1A和p16 mRNA在非小细胞肺癌(NSCLC)组织中表达的意义。方法采用RT—PCR检测96例NSCLC患者癌组织及相应的远癌正常肺组织中RASSF1A和p16 mRNA。结果①RASSF1A和p16 mRNA在远癌正常肺组织中均正常表达;在NSCLC组织中,RASSF1A mRNA表达下调或缺失率达53.12%(51/96).P16 mRNA达36.46%(35/96)。②RASSF1A mRNA下调或缺失与NSCLC病理分期和淋巴结转移有关。⑧p16 mRNA表达下调或缺失率女性(52.38%)高于男性(24.07%),50岁以上组(44.93%)高于50岁以下组(14.81%),腺癌(48.15%)高于鳞癌(21.43%),Ⅲ~Ⅳ期NSCLC下调或缺失率(45.61%)高于Ⅰ~Ⅱ期(23.08%),有淋巴结转移组(50.00%)高于无淋巴结转移组(13.89%),P均<0.05。④NSCLC组织中RASSF1A和p16 mRNA表达呈正相关(ra=0.278,P<0.05)。结论RASSF1A和p16 mRNA在NSCLC中表达下调或缺失率较高,二者可能参与调控NSCLC的发生和发展。

英文摘要:

[Objective] To explore the trans-crlptional expression of Ras association domain family 1A gene (RASSF1A) and p16 mRNA in non-small cell lung cancer (NSCLC) tissues. [Methods] RT-PCR was used to detect the expression of RASSF1A and p16 gene mRNA in 96 human NSCLC tissues and matched 96 normal lung tissues far from cancer area. [Results] ①The transcriptional expression of RASSF1A and p16 gene was normal in all normal lung tissues far from cancer area but lower or loss in 53.12%(51/96) carcinoma tissues for RASSF1A and in 36.46% (35/96) carcinoma tissues for p16 gene. ②The rate of low or loss of RASSFIA mRNA was significantly higher in the patients in advanced stages (61.04 % ) and in the patients with positive lymph node metastasis (65%) than in those in early stage (41.03%, P 〈 0.05) and in those without lymph node metastasis (33.33%, P 〈 0. 05). ③The rate of low or loss of p16 gene mRNA was remarkably higher in female group ( 52.38 % ), more than fifty years group (44.93 % ), adenocarcinoma group (48. 15 % ), stage Ⅲ and Ⅳ tumors (45.61%) and lymph node metastasis group (50.00%) than that in male group(24.07%),less than fifty years group (14.81%),squamous cell carcinoma group (21.83%),stage Ⅰ and Ⅱ tumors(23.08%) and non-lymph node metastasis group (13.89), P 〈 0. 05. ④The positive correlation between the transcriptional expression of RASSF1A and that of p16 gene was identified in 96 NSCLC tissues. [Conclusion] Loss or abnormal downregulation of RASSF1A and p16 mRNA is a frequent event in patients with NSCLS in Chlnese,which may play an important role in the malignant progress and prognosis of NSCLS.
