目的探讨WWOX蛋白、erbB2蛋白在非小细胞肺癌(NSCLC)组织中的表达及与肿瘤细胞增殖之间的关系。方法免疫组化方法检测50例鳞癌,31例腺癌及20例癌旁正常组织中WWOX蛋白、erbB2蛋白的表达,分析其与临床病理指标、肿瘤细胞增殖之间的关系。结果 WWOX蛋白在72.8%的NSCLC中失表达或表达降低,而相邻正常肺组织中有80.0%正常表达。WWOX蛋白的表达与组织类型、腺癌细胞分化程度密切相关(r=0.262,0.457,P〈0.05),在鳞癌和低分化腺癌中WWOX蛋白失表达或表达降低。肺癌中erbB2蛋白的过度表达与肺癌的临床分期、淋巴结转移相关,WWOX蛋白阳性表达与erbB2负相关(r=-0.239,P〈0.05),与肿瘤细胞增殖指数也呈负相关(r=-0.252,P〈0.05)。结论 WWOX蛋白的低表达与erbB2的过表达可以协同判断NSCLC的预后。
Objective To study the expressions of WWOX,erbB2 protein in non-small cell lung carcinoma (NSCLC).Methods WWOX,erbB2 protein expressions were evaluated by immunohistochemistry in 81 NSCLC patients (50 squamous cell carcinomas,31 adenocarcinomas) and 20 adjacent normal lung tissues and were correlated with histopathologic and clinical characteristics (histotype,grade,tumor-node-metastasis,stage,age,gender,index of cell proliferation Ki67).Results WWOX expression was absent or reduced in 72.8% of NSCLC,whereas it was normal in 80.0% of adjacent normal lung tissues.WWOX expression was strongly associated with tumor histology and histologic grade of adenocarcinoma (r=0.262,0.457,P0.05),WWOX protein expression was absent or strongly reduced in squamous cell carcinomas and poorly differentiated adenocarcinoma.The overexpression of erbB2 was associated with TNM stage and lymph node metastasis.Finally,WWOX expression was negatively related to the overexpression of erbB2 and the high index of cell proliferation (r=-0.239,-0.252,P0.05).Conclusions The low expression of WWOX protein and the overexpression of erbB2 protein are related to high aggressiveness of NSCLC.
