目的研究Zn^2+对人Ⅱ型肺泡上皮细胞炎性细胞因子IL-8和TNF-α mRNA和蛋白表达水平的影响。方法用含0、50、100和250μmol/L Zn^2+的F-12培养基培养人Ⅱ型肺泡上皮A549细胞,分别于3h和24h用RT-PCR半定量IL-8和TNF-α mRNA表达量,用RIA测定培养上清液中IL-8和TNF-α蛋白浓度。结果A549细胞染Zn^2+ 3h时A549细胞以浓度依赖方式高表达IL-8和TNF-α mRNA和蛋白;24h时IL-8和TNF-α蛋白表达量进一步增高,IL-8 mRNA表达下降,TNF-α mRNA继续保持高表达。结论 Zn^2+显著提高A549细胞IL-8和TNF-α表达量,Ⅱ型肺泡上皮细胞可能是Zn^2+导致的呼吸系统损伤中炎性细胞因子的重要来源。
Objective To study the effect of Zn^2+ on levels of the cytokines IL-8 and TNF-α mRNA expression of human Ⅱ alveolar epithelial cell, Methods Human Ⅱ alveolar epithelial A549 cells were treated with Zn^2+ at the doses of 0, 50, 100 and 250μmol/L Zn^2+ for 3 and 24 h, and the mRNA and protein level of IL-8 and TNF-α were determined by semiquantitive RT-RCR and RIA, respectively. Results Incubation of A549 cells with Zn^2+ for 3 h and 24h could stimulate the accumulations of IL-8 and TNF-α mRNA and protein of A549 cells in a concentration-dependent manner. Conclusion The production of IL-8 and TNF-α mRNA and protein from A549 cells stimulated by Zn^2+ could be important source of cytokines in respiratory damage induced by Zn^2+.
