目的:探讨非小细胞肺癌(NSCLC)组织中磷酸化AKT(p-AKT)和细胞增殖核抗原(PCNA)的表达。方法:应用免疫组化法检测80例NSCLC组织和35例非肿瘤性肺组织标本中p-AKT蛋白和PCNA的表达,分析p-AKT蛋白表达与NSCLC患者临床病理因素的关系。结果:非肿瘤性肺组织中p-AKT蛋白不表达,在NSCLC组织中的阳性表达率为78.8%(63/80),2组比较差异有统计学意义(P〈0.05);p-AKT蛋白的表达与患者年龄、性别、组织学类型及鳞癌的分化程度、TNM分期、淋巴结转移无关(P〉0.05),仅在高中分化腺癌组织中的阳性表达率(95.0%(19/20))高于低分化腺癌组织中的阳性表达率(50.0%(8/16),P〈0.05)。在NSCLC和非肿瘤性肺组织中,PCNA标记指数分别为(52.45±26.14)%和(9.57±2.20)%,2者相比差异有统计学意义(P〈0.05)。NSCLC组织中p-AKT蛋白的表达与PCNA呈正相关(r=0.376,P〈0.05)。结论:在NSCLC中存在AKT的活化,p-AKT的表达与肿瘤的高增殖活性有关。
Aim: To investigate the expression of phosphorylated AKT(p-AKT) and proliferating cell nuclear antigen (PCNA) in human non-small cell lung cancer (NSCLC) tissue and their correlations. Methods:The expression of p-AKT and PCNA in 80 cases of NSCLC and 35 cases of non-cancerous lung tissue were assessed by immunohistoehemistry, and their correlations with eliniopathological factors were analyzed. Results: The positive rate of p-AKT in lung cancer tissue ( 78.8 % ) was significantly higher than that of p-AKT in non-cancerous lung tissue (0.0%) ( P 〈 0. 05 ) ; the expression of p-AKT did not relate to age, sex, histologie subtype, squamous cancer differentiation, lymph node metastasis or TNM stages(P 〉0.05 ). The positive rate of p-AKT in well-differentiated and moderately-differentiated adenoeareinoma tissue (95.0%) was significantly higher than that in poorly-differentiated adenoeareinoma tissue (50.0%) , P 〈 0. 05. PCNA labeling indexes in NSCLC and non-cancerous lung tissue were (52.45 ± 26.14) % and (9.57 ± 2.20) % , P 〈 0. 05. In NSCLC tissue, the expression of p-AKT was positively correlated with PCNA ( r = 0. 376, P 〈 0. 05 ). Conclusion : AKT activation may be present in NSCLC. The expression of p-AKT is related to the high proliferating activity of tumor cells.
