癌基因AKT2在肺鳞癌和腺癌中的表达及与细胞增殖转移的关系

目的:探讨癌基因AKT2在肺鳞癌和腺癌中的表达及其与肿瘤细胞增殖与转移的关系。方法:应用免疫组化方法。检测肺鳞癌和腺癌组织80例、非肿瘤性肺组织标本35例中AKT2蛋白、PCNA的表达,并与临床病理因素进行相关性分析。结果:AKT2蛋白在肺鳞癌和腺癌组织中总阳性表达率为91.3%(73/80),显著高于在非肿瘤性肺组织中的表达5.7%(2/35)(P〈0.05);AKT2蛋白在淋巴结转移组和无淋巴结转移组阳性表达率分别为100%、80.6%,两组比较差异有统计学意义(P〈0.05);AKT2的表达与患者年龄、性别、组织学类型、组织分化及TNM分期均无关(P〉0.05)。在肺癌(鳞癌+腺癌)和非肿瘤性肺组织中,PCNA标记指数分别为(52.45±26.14)%和(9.57±2.20)%,两组之间差别有显著性意义(P〈0.01)。在肺鳞癌和腺癌中,AKT2蛋白的总表达与PCNA高增殖指数呈正相关关系(P〈0.01)。结论:AKT2的高表达与肺鳞癌和腺癌的高增殖活性及恶性行为有关。在肺癌的发生、发展中可能具有重要作用.

英文摘要:

Objective: To investigate the expression of oncogene AKT2 product, in human lung squamous cancer and adenocarcinoma tissue and its con'elation with cell proliferation and metastasis. Methods: The expression of AKT2 protein and proliferating cell nuclear antigen (PCNA) in 80 cases of lung squamous cancer and adenocarcinoma tissue and 35 cases of non-cancerous lung disease were assessed using immunohistochemistry, and their correlations with clinicopathologic factors were statistically analyzed. Results: The total positive rate of AKT2 in lung squamous cancer and adenocarcinoma tissue (91.3%)was significantly higher than that of AKT2 in no-cancerous lung disease (5.7%)(P〈0.05); In the group with and without lymph node metastasis. The positive rate was 100.0% and 80.6% respectively, the difference between them was significant (P〈0.05). No significant correlation was found between positive AKT2 expression and age, gender, histologic type and tumor differentiation and TNM staging (P〉0.05). PCNA labeling indexes in lung cancer tissue and non-cancerous lung disease was (52.45±26.14)% and (9.57±2.20)% (P〈0.01) respectively. In lung squamous cancer and adenocarcinoma, total express of AKT2 protein positively correlated with PCNA high proliferation index (P〈0.01).Conclusion: High expression of AKT2 may be related to the high proliferating activity and malignant behavior of lung squamous cancer and adenocarcinoma. AKT2 might play an important role in genesis and development of Lung cancer.
