娟珊牛是一个小型乳用牛品种,具有乳成分高、耐热性强、适应性好等优点。近年来,我国娟珊牛饲养量及进口规模逐渐增大。JH1是新近在娟珊牛中发现的一种隐性遗传缺陷,一旦纯合,将导致胚胎早期死亡,母牛流产。本研究首先建立了一种基于PCR-RFLP技术的JH1隐性致死突变的分子检测方法,进而对随机挑选的8头国内娟珊种公牛进行筛查,发现1头携带者。建议今后对进口娟珊牛进行严格的系谱分析及基因检测,从而降低引入JH1遗传缺陷基因的风险;同时对入围国家奶牛良种补贴项目的娟珊种公牛,应检测JH1基因型并在系谱中明确标注,以便实施科学选配。
Jersey cattle is a small body type dairy breed with high milk ingredients, good heat resistance and adaptability. In recent years, the breeding amount and imported scale of Jersey cattle was gradually increasing. JH1 is one recessive disorder which had been new found in Jersey cattle, the homozygous will lead to early embryonic death and abortion. Polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) method was established in this study to detect the alleles of JH1 among 8 Jersey bulls. As a result, one carrier was identiifed. Based on this, in our country, it is necessary to conduct strict pedigree analysis and genetic testing to reduce the risk of JH1 genetic defects in imported Jersey cattle. In order to implement a scientiifc selection and matching, the recessive alleles in Jersey cattle which have entered the Chinese national genetic improvement programs should be detected and conduct genealogy marked.
