采用乳酸提取法从克氏原螯虾虾壳中提取乳酸钙,在单因素试验的基础上,选择乳酸添加量、反应温度、反应时间进行Box-Behnken试验设计,采用响应面法分析这3个因素对响应值(钙提取量)的影响,从而对虾壳中钙的提取工艺进行优化.结果表明:最优乳酸钙提取条件是10 g虾壳的乳酸添加量为21.43 mL,温度61.34℃,时间40 min.此条件下提取量为765.12 mg,实测出的提取量为762.48 mg,两者基本相符,说明回归方程与实际情况拟合较好.
The paper extracted calcium lactate from Procambarus clarkia shell by using lactic acid. Based on the single factor experiments,a three-factor and three-level Box-Behnken central composite design was conducted to study the lactic acid amount,reaction temperature and reaction time on the response value(calcium extraction amount) in combination with response surface analysis so as to optimize the calcium extraction process. The results showed that the optimum extraction conditions were as follows: calculated by 10 g shrimp shell,lactic acid amount 21.43 mL,temperature 61.34 ℃ and extraction time 40 minutes; and the calcium extraction amount was 765.12 mg which was substantially accordant to the actual value 762.48 mg. The results indicated that the regression equation has good fitting effect.
