通过研究公、母鸽肉质性状候选基因肌肉抑制素(Myostatin,MSTN)在不同组织、不同发育阶段的表达变化规律,为研究鸽肉质性状的遗传调控机理提供依据。采用荧光探针RT—PCR,定量分析MSTN mRNA在公、母鸽的多个组织、多个发育时间点的表达量。RT—PCR结果表明,MSTNmRNA表达量在3个生理时期表现出先降低后升高的变化趋势,母鸽峰值出现较早。公鸽MSTN mRNA表达量最高的组织为肾脏,睾丸中表达丰度最低,青年期肝脏、心KMSTN mRNA表达量极显著高于其它时期的表达量,但在睾丸中没有检测到基因的表达。对不同生理时期母鸽MSTN mRNA的表达量比较后发现,青年期MSTN mRNA表达最高,显著高于其它时期。MSTN mRNA表达量受到组织、发育阶段和品种影响,该基因的表达在物种之间存在差异。
The tissue distribution and developmental specific expression of Myostatin (MSTN) mRNA transcripts in pigeon were studied in order to understand the genetic mechanism of meat quality traits. The expression of MSTN mRNA in different tissues and different development phases were revealed by QRT-PCR. The expression of MSTN mRNA decreased and then increased in 3 physiological periods of pigeon,and the peak of expression occurred earlier in female than in male. The highest level of expression occurred in the kidney and the lowest were in testis of male pigeon, respectively. The MSTN mRNA expression of male and female pigeon during the juvenescent phase were significantly higher than those during broodiness and incubation periods,and were not detectable in testis. These resuhs indicate that the expression of MSTN mRNA were influenced by tissue,developmental phases and breed factors.
