目的:探讨左金丸对人结肠癌多药耐药细胞HCT-116/L-OHP的逆转作用及其可能机制。方法:人结肠癌HCT-116细胞和HCT-116/L-OHP多药耐药细胞常规培养,左金丸干预48h后,MTT法检测HCT-116/L-OHP细胞对化疗药敏感性的影响,电感耦合等离子质谱法(ICP-MS)检测耐药细胞内奥沙利铂(L-OHP)药物浓度的改变,流式细胞仪分析细胞凋亡情况。结果:左金丸增加HCT-116/L-OHP细胞对奥沙利铂(L-OHP)、顺铂(DDP)、五氟尿嘧啶(5-Fu)、丝裂霉素C(MMC)4种化疗药物的敏感性,4种化疗药的半数抑制浓度(IC50)分别从(157.48±16.73)μg/mL下降至U(71.40±6.48)μg/mL,(26.38±1.92)μg/mL下降到(16.12±1.60)μg/mL,(33.29±3.64)μg/mL下降到(13.11±2.65)μg/mL,(12.50±2.49)μg/mL下降到(2.71±1.10)μg/mL;左金丸干预HCT-116/L-0HP细胞48h后细胞内L-OHP浓度明显增高(P〈0.01),耐药细胞凋亡率显著增加(P〈O.01)。结论:左金丸通过增加结肠癌多药耐药细胞内化疗药物浓度,抑制细胞增殖,诱导其凋亡,逆转HCT-116/L-OHP细胞株的多药耐药性。
Objective: To investigate the mechanisms and reversal effects of Zuojin Pills on human colon carcinoma multidrug resistance (MDR) in HCT-116/L-OHP cells. Methods: Human colon carcinoma MDR cells HCT-116 and HCT-116/ L-OHP were cultured conventionally. Zuojin Pills were used to intervene them. After 48 hours, the sensitivity to chemotherapy drugs and the reversal effect of multidrug resistance (MDR) in HCT-116/L-OHP cells was evaluated by the MTT assay. The variation of intracellular concentration of L-OHP in HCT-116/L-OHP cells was evaluated by the ICP-MS, and the effects of ZJW on L-OHP induced apoptosis and cell cycle were evaluated by flow cytometry (FCM). Results: Zuojin Pills could increase the sensitivity of HCT-116/L-OHP cells to L-OHP, DDP, 5-Fu, MMC. The IC50 value of L-OHP decreased from (157.48±16.73)μg/mL to (71.40±6.48)μg/mL, DDP was from (26.38±1.92)μg/mL to (16.12±1.60)μg/mL, 5-Fu was from (33.29±3.64)μg/mL to (13.1 1±2,65)μg/mL, and MMC was from (12.50±2.49)μg/mL to (2.71±1.10)μg/mL. Zuojin Pills increased the concentration of L-OHP in HCT-116/L-OHP cells after treatment for 48h (P〈0.01). The apoptosis rate of HCT-116/L-OHP cells increased (P〈0.01). Conclusion: Zuojin Pills reversed MDR in HCT-116/L-OHP cell, which possibly through inhibiting cell proliferation, inducing cell-apoptosis, ultimately affecting the concentration of drugs inside the cells.
