目的检测O^6-甲基鸟嘌呤-DNA甲基转移酶(O^6-methylguanine-DNA methyltransferase,MGMT)基因启动子区甲基化与维吾尔族子宫颈鳞癌之间的相关性。方法采用甲基化特异性聚合酶链反应(methylation specific PCR,MSP)对41例维吾尔族妇女子宫颈鳞癌组织及32名正常维吾尔族妇女子宫颈组织进行甲基化检测。结果32名正常子宫颈组织MGMT基因启动子区均未发生甲基化,为非甲基化状态;41例子宫颈鳞癌组织中共有13例发生MGMT基因启动子区甲基化(32%)。结论MGMT基因启动子区甲基化可能部分参与维吾尔族妇女子宫颈癌的发生发展过程,是维吾尔族子宫颈鳞癌发生过程中常见的分子事件,有可能作为维吾尔族妇女子宫颈癌诊断的肿瘤标志物。
Objective To detect the relationship between the promoter hypermethylation of O^6-methylguanine-DNA methyltransferase(MGMT) gene and cervical squamous cell carcinoma in Uigur women.Methods Forty-one tumor tissues of cervical squamous cell carcinoma and 32 normal cervical tissues were detected with methylation specific PCR(MSP) in Uigur women.Results No hypermethylation of MGMT promoter was detected in all of the normal cervical tissues,but 13 were detected in 41 cervical squamous cell carcinomas tissues(32%).Conclusion Hypermethylation of MGMT promoter possibly involves in the tumorigenesis and is a frequent molecular event in cervical squamous cell carcinoma.Hypermethylation of MGMT promoter can be a potential tumor marker of cervical squamous cell carcinoma in Uigur women.
