开发了一种共振拉曼光谱(RR)技术用于二价铜离子的定量评价的新方法.通常二价铜离子(Cu~(2+))在碱性的条件下能够被蛋白质还原成一价铜离子,而两个BCA探针分子能够与一价铜离子发生螯合反应,形成BCA/Cu~+复合物,它有很强的RR活性.因此,采用这一RR方法进行蛋白质检测,能够比传统方法的检测浓度范围更宽.并且,基于RR方法可以对Cu~(2+)的最低检测浓度达到5×10~(-7)mol/L,要比传统的比色和紫外可见吸收光谱法更灵敏.该方法提供了一种新颖且高度敏感的RR技术用于金属离子的定量研究.
In this paper,we have proposed a rapid and highly sensitive Bicinchoninic Acid (BCA) reagent-based Cu2+ ion quantitation tool using competitive resonance raman (RR) method.A chelation reaction between BCA and Cu+ which is reduced by protein in an alkaline environment.Two BCA molecules complex with a Cu+ create a BCA/Cu+ complex that has strong RR activity.The proposed RR method exhibit a much wider linear concentration range compared to UV -based methods. The detection limits based on the proposed RR method is as low as 5 x 10-7 molJL , which is lower than the traditional method. This novel and sensitive method exhibited an excellent application in quantitative determination of metal ions.
