建立了一种柱前衍生高效液相色谱法同时测定海带中岩藻聚糖的单糖及糖醛酸组成的方法。岩藻聚糖经4 mol/L三氟乙酸降解后,用1-苯基-3-甲基-5-吡唑啉酮(PMP)进行衍生化,并采用配有紫外检测器的反相高效液相色谱仪在250 nm波长下进行测定,实现了7种单糖和糖醛酸的良好分离。结果表明:该岩藻聚糖样品由5种单糖和2种糖醛酸组成,其物质量之比为甘露糖∶鼠李糖∶葡萄糖醛酸∶半乳糖醛酸∶葡萄糖∶半乳糖∶岩藻糖=0.03∶3.7∶0.22∶0.74∶0.12∶1.1∶4.1。该方法重现性良好,能够快速对多糖样品进行组成分析。
High performance liquid chromatography(HPLC) was employed for the analysis of monosaccharide and uronic acids in fucoidon from fresh seaweed with pre-column derivatization.After degradated by 4 mol/L trifluoroacetic acid and derivatived with 1-phenyl-3-methyl-5-pyrazolone(PMP),monosaccharides and uronic acids in fucoidon were separated by reversed-phase HPLC with gradient elution,and monitored by ultraviolet detector at 250 nm.The result show that fucoidon consists of 5 kinds of monosaccharides and 2 kinds of uronic acids.The ratio of mannose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactoses,fucose is 0.03∶3.7∶0.22∶0.74∶0.12∶1.1∶ 4.1.The developed method is suitable for the rapid analysis of polysaccharide with good reproducibility.
