利用选择性筛选方案从啤酒厂附近土壤样品中获得两株耐酸、耐热普鲁兰酶产生菌(PPL3和PPL4),16SrRNA基因系统发育分析结果显示,PPL3和PPIA-均为芽孢杆菌。酶学性质分析结果显示,最适pH值和最适温度均为7.O和90℃,粗酶液在pH值为5.0和90℃的条件下放置10h后残余酶活仍分别保持存85%和80%上,对酸和热表现出很好的稳定性。
Two acid- and thermo-tolerance pullulanase-producing strains (PPL3 and PPL4) were isolated by selective medium from soils collected near the brewery. 16S rRNA genes were PCR amplified, sequenced and phylogenetic analyzed, arid the results showed that the strains PPL3 and PPL4 were some species of Bacillus genus. The optimum pH and temperature were 7.0 and 90℃, respectively; above 85% and 80% activity of pullulanse from PPL3 were retained when crude enzyme in pH 5.0 buffer and on 90℃ for 10 h, respectively. The pullulanse from PPL3 and PPL4 exhibited the stability in acid and thermo conditions.
