目的:探讨三棱散水提物对人胃癌SGC-7901细胞增殖及凋亡影响及其可能机制。方法:制备三棱散水提物,培养人胃癌SGC-7901细胞,MTT法观察不同时间(24 h,72 h)不同浓度三棱散水提物对人胃癌SGC-7901细胞增殖的影响。用流式细胞仪检测三棱散水提物对细胞周期阻滞及其凋亡的影响。RT-PCR法检测不同浓度三棱散水提物作用人胃癌SGC-7901细胞72 h后细胞NF-κBp65 mRNA、cyclin D1 mRNA及p16 mRNA表达情况。结果:随着三棱散水提物浓度增高和作用时间的延长,人胃癌SGC-7901细胞增殖抑制率逐渐增高(P〈0.05),同一浓度不同时间的细胞增殖抑制率相比,有显著性差异(P〈0.05)。人胃癌SGC-7901细胞在G0/G1期表现出明显的增加,S期细胞明显的减少,各浓度组均明显诱导细胞凋亡(P〈0.05),其凋亡率随三棱散浓度增加而提高。RT-PCR法检测显示,随着三棱散水提物浓度的增加,NF-κBp65 mRNA、cyclin D1 mRNA表达逐渐减少,p16 mRNA表达逐渐增加。三棱散水提物浓度5.0 mg/m L,10.0 mg/m L时,NF-κBp65 mRNA、cyclin D1 mRNA及p16 mRNA表达存在显著性差异(P〈0.05或P〈0.01)。结论:三棱散水提物对人胃癌SGC-7901细胞有抑制增殖和促凋亡作用,呈时间和浓度依赖性,且G0/G1期细胞数量增多,S期细胞数量减少。RT-PCR法检测显示,NF-κBp65 mRNA、cyclin D1 mRNA呈现低表达,p16 mRNA呈现高表达,由此推测,NF-κBp65表达下调会直接影响Cyclin D1表达,发挥细胞周期的正反馈调节,p16表达上调可启动细胞周期的负反馈调节,共同作用产生细胞增殖抑制及促进凋亡的作用。
Objective: To investigate the effects of Sanleng Powder extract( SLSE) on proliferation and apoptosis of the human gastric carcinoma cell line 7901 and its possible mechanism. Methods: The growth inhibition effect of SLSE on human gastric carcinoma cell line 7901 was observed by using MTT assay and cell apoptosis was analyzed by flow cytometry. The expressions of NF-κBp65 mRNA,cyclin D1 mRNA and p16 mRNA in SGC- 7901 cells were analyzed by reverse transcription- polymerase chain reaction( RT- PCR). Results: The SGC- 7901 cells were treated with various concentrations of SLSE for 48 h and 72 h and cell viability was determined by Alamar Blue assay. Cell growth was suppressed by SLSE of different concerntrions( 0,1. 25 mg / m L,2. 5 mg / m L,5. 0 mg / m L,10. 0 mg / m L). SLSE inhibited the growth of gastric cancer cells in a dose- dependent manner. The cells were treated with various concentrations( 0,2. 5 mg / m L,5. 0 mg / m L,10. 0 mg / m L) of SLSEs for 72 h. The results showed that SLSEs were capable of inducing an increase in the percentages of G1- phase cells and the number of apoptotic cells( P〈0. 05 or P〈0. 01). Real- time quantitative PCR was used to detect the mRNA expressions of NF- κBp65,cyclin D1 and p16 at72 h after SLSE treatment. The results revealed a significant down- regulation of mRNA expressions of NF- κBp65 and cyclin D1 and up- regulation of the mRNA expression levels of p16 as compared with those of the controls. Conclusion: The results show that SLSE has significant action of anti- proliferation and pro- apoptosis on human gastric carcinoma cell line 7901. The down- regulation of NF- κBp65 mRNA expression will directly affect the expression of Cyclin D1 mRNA which suppresses the cell proliferation inhibition and promotes the cell apoptosis. The p16 mRNA showed high expression,which can activate the negative feedback of cell cycle. The results suggest that SLSE may have an effect on the pathway of gastric cancer genesis.
