对中国境内19个地区的东方蜜蜂的EF-1α基因进行了测序,并结合蜜蜂属内其他蜂种以及其他地区东方蜜蜂的相应序列信息,分析中国境内东方蜜蜂的遗传分化状况.测序后得到了中国境内19个不同地区的EF-1α部分序列,将获得基因序列提交至GenBank,登录号为:KF663 570~KF663 573.通过对19个地区的东方蜜蜂EF-1α基因序列比对后发现,这些序列间的相似性均大于99.3%,序列中共有3个变异位点.基于EF-1α构建的系统发育树显示19个地区的东方蜜蜂与印度尼西亚、斯里兰卡和马来西亚的东方蜜蜂,以及沙巴蜂和绿努蜂聚为一个分支.研究的结果表明,该基因不能够用于东方蜜蜂的种内分化研究,但作为蜜蜂种间的系统发育研究确是一种很好的分子标记.
The EF-1agene of Apis ceranafrom 19 regions in China were sequenced and the genetic differentiation status of A.ceranain China was analyzed with the corresponding sequence information of other bee species.The sequences of EF1-αin 19 different regions in China were submitted to GenBank after sequencing.The accession number was:KF663570~KF663573.Through the comparison of 19 regions of the EF-1alpha gene sequence of A.cerana,it was found that the similarity between these sequences was more than 99.3%,and there were 3mutation sites in the sequence.The phylogenetic tree constructed basing on the EF1-αgene shows that the A.ceranain 19 regions with A.ceranain Indonesia,A.ceranain Sri Lanka,and A.ceranain Malaysia,Apis nuluensis and Apis koschevnikovi were in one cluster.The results of this study showed that EF1-αgene could be used for a molecular markes of phylogenetic analysis among species rather than among subspecies.
