回药失荅剌知丸抗大鼠脑缺血再灌注后细胞凋亡的实验研究

目的:观察回药失荅剌知丸对大鼠脑缺血再灌注后脑组织中Fas及Fas L表达的影响,探讨失荅剌知丸治疗脑缺血性疾病的机制。方法:SD雄性大鼠随机分为假手术组、模型组、金纳多组、失荅剌知丸低浓度组、失荅剌知丸中浓度组、失荅剌知丸高浓度组,局灶性脑缺血再灌注模型制备成功后分别于12h、24h、72h取材,采用干湿重法测脑含水量,免疫荧光法观察Fas阳性细胞表达量,TUNEL和免疫组化法观察神经细胞凋亡情况及Fas L表达的变化。结果:与假手术组相比,缺血再灌注损伤后脑含水量、神经细胞凋亡数量及Fas、Fas L的表达明显增高(P〈0.01,P〈0.05);与模型组比较,用药组各时段脑含水量、神经细胞凋亡数量及Fas、Fas L的表达均减弱(P〈0.05),以失荅剌知丸高浓度组的表达减弱显著(P〈0.01)。结论:失荅剌知丸能够有效减轻脑组织水肿,抑制Fas、Fas L的表达,其发挥神经保护的作用机制可能与抑制神经细胞凋亡有关。

英文摘要:

Objective : To observe the effect of expression of Fas and FasL in the hippocampus of ischemia - reperfusion injury rats brain tissue by taking Shida Lazhi Wan, and to explore Shida Lazhi Wan mechanism for the treatment of cerebral ischemic diseases. Methods: SD male rats were randomly divided into 6 groups: sham group, the ischemia reperfusion model group (hereinafter referred to as the model group), JinNaDuo group, Shida Lazhi Wan low concentration group, Shidalazhiwan medium concentration group, Shida Lazhi Wan high concentration group, After focal cerebral ischemia models were established, at 12h, 24h, 72h, neurons apop- tosis, wet and dry weight measured brain water content, immunofluo - rescence techniques to measure Fas expression, TUNEL and immunohistochemical method to observe neuronal apoptosis and the change of FasL expression. Result : To compared with the sham op- eration group, after ischemia - reperfusion injury , brain water, number of nerve cell apoptosis and Fas, FasL expression were signifi- cantly increased (P 〈0. 01, P 〈0. 05) ; compared with the model group, every treatment groups brain water number, nerve cell ap- optosis and Fas, FasL expression were recede ( P 〈 0. 05 ), especially Shida Lazhi Wan high concentration group Reduce significantly (P 〈 0. 01 ). Conclusion: Shida Lazhi Wan can .7 reduce brain water effectively, inhibition of Fas and FasL express, its nerve protec- tion mechanism may be related to inhibition of neural cell apoptosis.
