目的:探讨流式细胞术微球阵列法(CBA)在细胞因子检测中的应用及血清肿瘤坏死因子(TNF)、白细胞介素6(IL-6)和白细胞介素10(IL-10)在不同亚型急性白血病(AL)中的表达水平。方法:采用BD CBA Flex Set IL-6、IL-10、TNF试剂盒和流式细胞术检测29例急性白血病患者和20例正常人血清样本中3种细胞因子水平。结果:AL患者IL-6、IL-10和TNF水平显著高于正常对照组。急性淋巴细胞白血病(ALL)的IL-10和TNF水平较急性非淋巴细胞白血病(ANLL)有显著性提高(P〈0.05)。在ANLL亚型中,M1型IL-6水平最高。结论:CBA法能快速、准确地定量检测细胞因子;AL血清中IL-6、IL-10和TNF水平升高。可能与疾病的发生发展存在相关性。
Aim: To explore the application of cytometric bead array (CBA) method in detection of cytokines and detect the levels of IL-6,IL-10 and interferon (IFN) in the serum from patients with different subtypes of acute leukemia(AL). Methods: The levels of three cytokines (IFN, IL-6, and IL-10) in the serum from 29 cases of patients with AL and 20 cases of normal persons were detected by using flow cytometry and BD CBA Flex Set kit. Results: The levels of IFN, IL-6, and IL-10 in AL group were significantly higher than those in controls. Compared with acute non-lymphoid leukemia (ANLL) group, the levels of IFN and IL-10 in acute lymphoid leukemia (ALL) group were significantly higher (P 〈0.05). The level of IL-6 in M1 was the highest among the ANLL subtypes. Conclusion: Detecting cytokines by CBA method is quick and accurate; IFN, IL-6, and IL-10 might take part in the process of AL according to their increased levels in AL.
