白僵菌是蛀干害虫桑天牛的生物防治菌种。利用紫外线对前期筛选出的生防用白僵茵菌株Bb00进行诱变,筛选出几丁质酶活力高的突变株,再测定其对桑天牛幼虫的致病力,获得毒力高于出发菌株的突变株Bb00-3和Bb00-4。用这2个菌株的1×10^8mL-1孢子悬液喷施桑天牛幼虫,至处理第6天时的校正死亡率分别达86.75%和79.95%,致死中时(L疋。)分别为4.13d和4.46d,与出发菌株相比具有显著差异(P〈0.05)。突变株Bb00-3经继代培养6代,每代的产孢量始终高出相应代数出发菌株的1倍以上,6次继代过程中亦没有出现致病力的明显降低,表现出较好的遗传稳定性。
Beauveria bassiana is a fungal strain for biological control against mulberry stem borer, Apriona germari. Ul- traviolet was used to irradiate B. bassiana Bb00 strain, which had been obtained from our previous screening work, to create mutants with high chitinase activity. After determining their infectivity to A. germari larvae, two highly virulent mutational strains, named Bb00-3 and Bb00-4, were obtained. After their 1 x 108 mL-1 spore suspensions were used to treat A. germari larvae, the corrected mortality at the 6th day post-inoculation reached 86.75% and 79.95%, and the median lethal time (LT50) of Bb00-3 and Bb00-4 was 4.13 d and 4.46 d respectively, being significantly different with the original strain (P〈0.05). During 6 generations of sub-culture, spore yield of mutational strain Bb00-3 was always 1 times higher than that of the original strain. Moreover, the virulence of each generation was not obviously weakened, showing excellent genetic stability.
