目的:阐释葛根芩连汤治疗2,4,6-三硝基本磺酸(TNBS)+乙醇诱导的溃疡性结肠炎(UC)的作用与机制。方法:雄性SD大鼠60只,分为正常组,模型组,阳性药组(柳氮磺胺吡啶300mg·kg-1·d-1),葛根芩连汤高、中、低剂量组(分别予葛根芩连汤是28.05、18.15、8.25g·kg-1·d-1),每组10只,除正常组外,各组均以TNBS/乙醇混合液灌入大鼠结肠部位造溃疡性结肠炎模型,观察大鼠的一般情况及结肠组织损伤,检测大鼠血浆髓过氧化物酶(MPO),丙二醛(MDA),超氧化物歧化酶(SOD),过氧化氢(H2O2),P-选择素(P-selectin),白细胞介素4(IL-4),白细胞介素18(IL-18)含量。结果:葛根芩连汤能明显改善UC大鼠的一般状况,显著降低UC大鼠DAI指数、结肠组织损伤指数(P〈0.05,P〈0.01),减少充血和中性粒细胞浸润;葛根芩连汤能够降低血浆中MPO、H_2O_2、NO、MDA、P-selectin、IL-18的表达,以及提高血浆SOD、IL-4的表达。结论:葛根芩连汤对UC大鼠的结肠组织损伤具有明显的治疗作用,可能通过增加机体抗氧化作用,降低促炎因子的表达,从而达到治疗目的。
Objective: To explore mechanism of Gegen Qinlian Decoction(GGQLD) in rats with ulcerative colitis. Methods: Sixty male SD rats were divided into the control group, the model group, the sulfasalazine(300mg·kg-1·d-1) group, and the high, middle and low dose of GGQLD(28.05, 18.15g/kg, 8.25g·kg-1·d-1) groups, 10 rats of each group. Rats in all groups except for the control group received TNBS/ethanol mixture by gavage to induce the ulcerative colitis model. The general situations and damage of colon tissues were observed. The level of myeloperoxidase(MPO), malondialdehyde(MDA), superoxide dismutase(SOD), hydroperoxide(H_2O_2), P-selectin, interleukin-4 and interleukin-18 in plasma were detected with Enzyme-linked immunosorbent assay. Results: GGQLD significantly improved general conditions of UC rats. GGQLD significantly reduced DAI index and colon tissue injury index(P〈0.05, P〈0.01) of UC rats, as well as congestion and neutrophil infiltration. The high dose GGQLD group effectively inhibited the expression of MPO, H_2O_2, NO, MDA, P-selectin, IL-18, and increased the level of IL-4 and SOD in plasms. Conclusion: GGQLD could significantly improve the UC rats, which may be associated with the increase of body's antioxidant effect and the inhibition of proinflam matory factors.
