目的建立高效液相色谱电化学法测定血浆丙泊酚浓度,初步评价新型丙泊酚目标控制输注(TCI)系统性能。方法丙泊酚通过装有“Diprifusor’Tc2”系统的Alaris输液泵给药,应用Microsoft Excel 2000软件,分析10例病人血浆丙泊酚浓度的实测值与目标值的相关性。结果63份血浆样品丙泊酚实测浓度与目标浓度执行误差的中位数(MDPE)=-10.75%,执行误差绝对值的中位数(MDAPE)=8.57%,摆动度(MDAPDE)=14.60%。丙泊酚血药实测浓度与目标浓度的相关系数r=0.68。结论HPLC-ECD法测定血浆丙泊酚浓度,方法较简便、准确.灵敏度高.重现性好,适用于临床丙泊酚血药浓度监测及药动学研究。新型丙泊酚目标控制输注(TCI)系统偏离性较小。精密度高。但摆动度偏大.用于国人需优化。
OBJECTIVE To establish a HPLC-ECD method for determi propofol concentration in plasma, and to evaluate initially the performance of a new target-controlled infusion(TCI) system in Chinese. METHODS Propofol was infusioned by a Alaris pump contained diprifusor' TC2 system. The data were analyzed by Microsoft Excel 2000 program. RESULTS The median performance error (MDPE) was - 10.75 %, The median absolute performance error (MDAPE) was 8.57 %, The wobble (the median absolute deviation of each PE from the MDPE) was 14.60 %. The correl of two groups was 0.68. CONCLUSION HPLC-ECD is rapid, simple, accurate and sensitive to determinate propofol concentration in plasma. It is suitable for monitoring propofol and pharmacokinetics study. The performance of the TCI of propofol is acceptable for clinical use.but the propofol pharmacokinetics parameters adopted in the system are not fit for chinese and it may be required to be optimized.
